Williamson, W. T. (North Carolina State of the University of North Carolina, Raleigh), S. B. Tove, and M. L. Speck. Extracellular proteinase of Streptococcus lactis. J. Bacteriol. 87:49-53. 1964.-Streptococcus lactis was shown to produce an extracellular proteolytic enzyme(s). A 120-fold purification of the proteinase was obtained from the cell-free culture medium by ammonium sulfate fractionation, calcium phosphate gel treatment, and chromatography on diethylaminoethyl cellulose. Only 32% of the activity was destroyed by heating at 98 C for 1 hr. The requirement of free sulfhydryl groups for enzyme activity was indicated by inhibition with p-chloromercuric benzoate. Diisopropyl fluorophosphate did not inhibit the proteinase.

Plasmid distribution and evidence for a proteinase plasmid in Streptococcus lactis C2-1.
Five plasmids, distinguishable by their molecular weights (10(6), 2 x 10(6), 5 x 10(6), 10(7), 3 x 10(7), respectively) were isolated from Streptococcus lactis C2. A spontaneous proteinase-negative derivative of this strain lacked the 10(7) plasmid.

متن کاملDeletion analysis of the proteinase gene of Streptococcus cremoris Wg2.
The Streptococcus cremoris Wg2 proteinase gene, cloned in S. lactis, specified a proteinase which exhibited the same specificity toward casein as did the proteinase isolated from the original host. Although the cloned gene lacked the last 130 codons, the proteinase still specifically degraded beta-casein. Deletion of the C-terminal 343 amino acids from the proteinase did not influence this spec...
متن کاملAutolysis of Lactococcus lactis is influenced by proteolysis.
The autolysin AcmA of Lactococcus lactis was shown to be degraded by the extracellular lactococcal proteinase PrtP. Autolysis, as evidenced by reduction in optical density of a stationary-phase culture and concomitant release of intracellular proteins, was greatly reduced when L. lactis MG1363 cells expressed the cell wall-anchored lactococcal proteinase PrtP of the PI-type caseinolytic specifi...

Streptococcus thermophilus ATCC 19258, Lactobacillus delbrueckii subsp. bulgaricus T-11, and Lactococcus lactis subsp. lactis IL1403 were transformed with the alpha-amylase gene (amyA) from Streptococcus bovis 148 by using a wide host-range vector, and all the transformants secreted the alpha-amylase successfully. Since the promoter and the secretion signal of the amyA gene were functional in t...
